人chemerin elisa試劑盒 技術(shù)流程ELISA的基礎(chǔ)是抗原或抗體的固相化及抗原或抗體的酶標(biāo)記。結(jié)合在固相載體表面的抗原或抗體仍保持其免疫學(xué)活性,酶標(biāo)記的抗原或抗體既保留其免疫學(xué)活性,又保留酶的活性。在測(cè)定時(shí),受檢標(biāo)本(測(cè)定其中的抗體或抗原)與固相載體表面的抗原或抗體起反應(yīng)。用洗滌的方法使固相載體上形成的抗原抗體復(fù)合物與液體中的其他物質(zhì)分開(kāi)。再加入酶標(biāo)記的抗原或抗體,也通過(guò)反應(yīng)而結(jié)合在固相載體上。此時(shí)固相上的酶量與標(biāo)本中受檢物質(zhì)的量呈一定的比例。加入酶反應(yīng)的底物后,底物被酶催化成為有色產(chǎn)物,產(chǎn)物的量與標(biāo)本中受檢物質(zhì)的量直接相關(guān),故可根據(jù)呈色的深淺進(jìn)行定性或定量分析。由于酶的催化效率很高,間接地放大了免疫反應(yīng)的結(jié)果,使測(cè)定方法達(dá)到很高的敏感度。 ELISA可用于測(cè)定抗原,也可用于測(cè)定抗體。然而,影響Elisa試驗(yàn)結(jié)果的因素很多,故加強(qiáng)各個(gè)環(huán)節(jié)的質(zhì)量保證才能充分發(fā)揮其方法學(xué)的優(yōu)點(diǎn)。

使用方法1、 血清:操作過(guò)程中避免任何細(xì)胞刺激。使用不含熱原和內(nèi)毒素的試管。收集血液后,1000×g離心10分鐘將血紅細(xì)胞迅速小心地分離。2、 血漿:EDTA、檸檬酸鹽、肝素血漿可用于檢測(cè)。1000×g離心30分鐘去除顆粒。3、 細(xì)胞上清液:1000×g離心10分鐘去除顆粒和聚合物。4、 組織勻漿:將組織加入適量生理鹽水搗碎。1000×g離心10分鐘,取上清液。5、 保存:如果樣品不立即使用,應(yīng)將其分成小部分-70℃保存,避免反復(fù)冷凍。盡可能的不要使用溶血或高血脂血。如果血清中大量顆粒,檢測(cè)前先離心或過(guò)濾。不要在37℃或更高的溫度加熱解凍。應(yīng)在室溫下解凍并確保樣品均勻地充分解凍。操作步驟1:雙抗體夾心法1.包被:用0.05MPH9.牰碳酸鹽包被緩沖液將抗體稀釋至蛋白質(zhì)含量為1~10μg/ml。在每個(gè)聚苯乙烯板的反應(yīng)孔中加0.1ml,4℃ 過(guò)夜。次日,棄去孔內(nèi)溶液,用洗滌緩沖液洗3次,每次3分鐘。(簡(jiǎn)稱洗滌,下同)。2. 加樣:加一定稀釋的待檢樣品0.1ml于上述已包被之反應(yīng)孔中,置37℃ 孵育1小時(shí)。然后洗滌。(同時(shí)做空白孔,陰性對(duì)照孔及陽(yáng)性對(duì)照孔)。3. 加酶標(biāo)抗體:于各反應(yīng)孔中,加入新鮮稀釋的酶標(biāo)抗體(經(jīng)滴定后的稀釋度)0.1ml。37℃ 孵育0.5~1小時(shí),洗滌。4. 加底物液顯色:于各反應(yīng)孔中加入臨時(shí)配制的TMB底物溶液0.1ml,37℃ 10~30分鐘。5. 終止反應(yīng):于各反應(yīng)孔中加入2M硫酸0.05ml。6. 結(jié)果判定:可于白色背景上,直接用肉眼觀察結(jié)果:反應(yīng)孔內(nèi)顏色越深,陽(yáng)性程度越強(qiáng),陰性反應(yīng)為無(wú)色或極淺,依據(jù)所呈顏色的深淺,以“+”、“-”號(hào)表示。也可測(cè)OD值:在ELISA檢測(cè)儀上,于450nm(若以ABTS顯色,則410nm)處,以空白對(duì)照孔調(diào)零后測(cè)各孔OD值,若大于規(guī)定的陰性對(duì)照OD值的2.1倍,即為陽(yáng)性。操作步驟2:間接法1.用包被緩沖液將已知抗原稀釋至1~10μg/ml,每孔加0.1ml,4℃過(guò)夜;2.次日洗滌3次;3.加一定稀釋的待檢樣品(未知抗體)0.1ml于上述已包被之反應(yīng)孔中,置37℃孵育1小時(shí),洗滌;4.(同時(shí)做空白、陰性及陽(yáng)性孔對(duì)照)于反應(yīng)孔中,加入新鮮稀釋的酶標(biāo)第二抗體(抗抗體)0.1ml;5.37℃孵育30-60分鐘,洗滌;6.’最后一遍用DDW洗滌。其余步驟同“雙抗體夾心法”的4、5、6。
標(biāo)本要求1.標(biāo)本采集后盡早進(jìn)行提取,提取按相關(guān)文獻(xiàn)進(jìn)行,提取后應(yīng)盡快進(jìn)行實(shí)驗(yàn)。若不能馬上進(jìn)行試驗(yàn),可將標(biāo)本放于-20℃保存,但應(yīng)避免反復(fù)凍融2.不能檢測(cè)含NaN3 的樣品,因NaN3 抑制辣根過(guò)氧化物酶的(HRP)活性。

注意事項(xiàng)1.試劑盒從冷藏環(huán)境中取出應(yīng)在室溫平衡15-30 分鐘后方可使用,酶標(biāo)包被板開(kāi)封后如未用完,板條應(yīng)裝入密封袋中保存。2.濃洗滌液可能會(huì)有結(jié)晶析出,稀釋時(shí)可在水浴中加溫助溶,洗滌時(shí)不影響結(jié)果。3.各步加樣均應(yīng)使用加樣器,并經(jīng)常校對(duì)其準(zhǔn)確性,以避免試驗(yàn)誤差。一次加樣時(shí)間最好控制在5 分鐘內(nèi),如標(biāo)本數(shù)量多,推薦使用排槍加樣。4. 請(qǐng)每次測(cè)定的同時(shí)做標(biāo)準(zhǔn)曲線,最好做復(fù)孔。如標(biāo)本中待測(cè)物質(zhì)含量過(guò)高(樣本OD 值大于標(biāo)準(zhǔn)品孔第一孔的OD 值),請(qǐng)先用樣品稀釋液稀釋一定倍數(shù)(n 倍)后再測(cè)定,計(jì)算時(shí)請(qǐng)最后乘以總稀釋倍數(shù)(×n×5)。5. 封板膜只限一次性使用,以避免交叉污染。6.底物請(qǐng)避光保存。7.嚴(yán)格按照說(shuō)明書(shū)的操作進(jìn)行,試驗(yàn)結(jié)果判定必須以酶標(biāo)儀讀數(shù)為準(zhǔn).8.所有樣品,洗滌液和各種廢棄物都應(yīng)按傳染物處理。9.本試劑不同批號(hào)組分不得混用。10.保存條件及有效期:1.試劑盒保存:;2-8℃。2.有效期:6 個(gè)月

UNSATURATED SERIES Straight chain fatty acids, mono basic, aliphatic, terminating in a single carboxyl group and of reduced hydrogen content than their saturated counterparts, constitute the ethylenic family of unsaturated fatty acids. They are of considerable importance since linkage with other chemical radicals is readily accomplished at the double bond positions. Multiple unsaturation is found in almost all natural occurring vegetable and animal fats. Research in recent years has shown them to be of great significance. Double bonds are susceptible to oxidation so that stability is a factor to be reckoned with (especially polyunsaturated compounds) when researching unsaturated fatty acids or their corresponding esters. Highly purified unsaturated compounds prepared in our laboratory are always flushed several times with nitrogen, sealed in glass under vacuum and stored under cold conditions until ready for shipment. Nu-Chek suggests that when your shipment is received, they should be stored under cold conditions until ready for use. Once vial is opened protect with N2 and use as soon as possible.

GLYCERIDES The distribution of random fatty acids in naturally occurring fats and oils in the form of triglycerides dates back to antiquity. Indeed, the use of soap-cosmetics and oil lubricants dates back thousands of years. While we at Nu-Chek do not profess to go back quite that far, we would like to say that we have been synthesizing pure gylceride standards for the scientific community for over thirty years. The general procedure for all our triglyceride preparations is well exemplified in Nu-Chek's Triolein. Our starting material is chromatographically pure Methyl Oleate (isolated from the best virgin olive oil available) which is interesterified for the production of crude Triolein. To assure geometricstability of cis double bonds, the material is further purified by such physical techniques as low temperature crystallization and column chromatography. The final purity is followed both by gas liquid and thin layer chromatography. Other triglycerides are prepared in a similar manner commencing with the particular oil which contains the best concentrates of the specific ester required.
